RESUMO

Spinal cord injury (SCI) constitutes a significant clinical challenge, and there is extensive research focused on identifying molecular activities that can facilitate the repair of spinal cord injuries. Mammalian sterile 20-like kinase 2 (MST2), a core component of the Hippo signaling pathway, plays a key role in apoptosis and cell growth. However, its role in neurite outgrowth after spinal cord injury remains unknown. Through comprehensive in vitro and in vivo experiments, we demonstrated that MST2, predominantly expressed in neurons, actively participated in the natural development of the CNS. Post-SCI, MST2 expression significantly increased, indicating its activation and potential role in the early stages of neural recovery. Detailed analyses showed that MST2 knockdown impaired neurite outgrowth and motor function recovery, whereas MST2 overexpression led to the opposite effects, underscoring MST2's neuroprotective role in enhancing neural repair. Further, we elucidated the mechanism underlying MST2's action, revealing its interaction with AKT and positive regulation of AKT activity, a well-established promoter of neurite outgrowth. Notably, MST2's promotion of neurite outgrowth and motor functional recovery was diminished by AKT inhibitors, highlighting the dependency of MST2's neuroprotective effects on AKT signaling. In conclusion, our findings affirmed MST2's pivotal role in fostering neuronal neurite outgrowth and facilitating functional recovery after SCI, mediated through its positive modulation of AKT activity. In conclusion, our findings confirmed MST2's crucial role in neural protection, promoting neurite outgrowth and functional recovery after SCI through positive AKT activity modulation. These results position MST2 as a potential therapeutic target for SCI, offering new insights into strategies for enhancing neuroregeneration and functional restoration.

RESUMO

The discovery of promising cytokines and clarification of their immunological mechanisms in controlling the intracellular fate of Mycobacterium tuberculosis (Mtb) are necessary to identify effective diagnostic biomarkers and therapeutic targets. To escape immune clearance, Mtb can manipulate and inhibit the normal host process of phagosome maturation. Phagosome maturation arrest by Mtb involves multiple effectors and much remains unknown about this important aspect of Mtb pathogenesis. In this study, we found that interleukin 16 (IL-16) is elevated in the serum samples of Tuberculosis (TB) patients and can serve as a specific target for treatment TB. There was a significant difference in IL-16 levels among active TB, latent TB infection (LTBI), and non-TB patients. This study first revealed that macrophages are the major source of IL-16 production in response to Mtb infection, and elucidated that IL-16 can promote Mtb intracellular survival by inhibiting phagosome maturation and suppressing the expression of Rev-erbα which can inhibit IL-10 secretion. The experiments using zebrafish larvae infected with M. marinum and mice challenged with H37Rv demonstrated that reducing IL-16 levels resulted in less severe pathology and improved survival, respectively. In conclusion, this study provided direct evidence that Mtb hijacks the host macrophages-derived interleukin 16 to enhance intracellular growth. It is suggesting the immunosuppressive role of IL-16 during Mtb infection, supporting IL-16 as a promising therapeutic target.

RESUMO

Temperature fluctuations occurring during the cold chain logistics of salmon contribute to lipid oxidation. This study aimed to simulate cold chain interruption through freeze-thaw operations and evaluate the lipidomics data from salmon samples subjected to different numbers of freeze-thaw cycles by using rapid evaporative ionization mass spectrometry (REIMS) combined with an intelligent surgical knife (iKnife). The results indicated significant differences in the relative abundance of characteristic ions among the samples (p < 0.05). A total of 34 ions with variable importance for the projection values ≥1 were identified as potential biomarkers, including m/z 719.4233 ([PCC36:5-NH(CH3)3]-), m/z 337.3134 ([FAC22:1]-), m/z 720.4666 ([PEC35:6-H]-), m/z 309.2780 ([FAC20:1]-), m/z 777.4985 ([PCC40:4-NH(CH3)3]-), m/z 745.4421 ([PCC38:6-NH(CH3)3]-/[PEC38:6-NH3]-), m/z 747.4665 ([PCC38:5-NH(CH3)3]-/[PEC38:5-NH3]-), etc. The degree of lipid oxidation was found to be associated with the number of freeze-thaw cycles, exhibiting the most significant alterations in the relative abundance of lipid ions in the 8T samples. Additionally, sensory evaluation by the CIE-L*a*b* method and volatile analysis by headspace solid-phase microextraction gas chromatography-mass spectrometry demonstrated significant differences (p < 0.05) in color and odor among the salmon samples, with a correlation to the number of freeze-thaw cycles. The primary compounds responsible for alterations in salmon odor were aldehydes with lower odor thresholds. In summary, the iKnife-REIMS method accurately differentiated salmon muscle tissues based on varying levels of lipid oxidation, thus expanding the application of REIMS.

RESUMO

As the "core" of fertilizer machinery, the fertilizer apparatus has a significant impact on the effect of fertilization operation. At present, the straight-grooves type external groove wheel fertilizer apparatus is widely used, which has the problem that the fertilizer flow fluctuates periodically and is not conducive to precision fertilization. Therefore a kind of the spiral groove wheel fertilizer apparatus is proposed in the paper, and the designed fertilizer apparatus is experimented and optimized by using Design-Expert Software 10 and a self-made fertilizer experiment bench. Taking the fertilization amount and the variation coefficient of fertilization amount as the experiment index, the interaction effects of the groove radius, spiral rising angle, groove wheel rotation speed, and fertilizer tongue inclination of the spiral groove wheel type fertilizer wheel on the fertilization performance are analyzed. The quadratic regression models of multiple factors of the fertilization amount and the variation coefficient of fertilization amount are established. Through optimization and experiment validation, the optimal combination of parameters is determined as follows: the groove radius is 13.5 mm, the spiral rising angle is 62°, the groove wheel rotation speed is 29.4 r/min, the fertilizer tongue inclination is 5°, the fertilization amount is 17.810 g and the variation coefficient of fertilization amount is 5.60%.

RESUMO

Recently, aqueous zinc-ion batteries with conversion mechanisms have received wide attention in energy storage systems on account of excellent specific capacity, high power density, and energy density. Unfortunately, some characteristics of cathode material, zinc anode, and electrolyte still limit the development of aqueous zinc-ion batteries possessing conversion mechanism. Consequently, this paper provides a detailed summary of the development for numerous aqueous zinc-based batteries: zinc-sulfur (Zn-S) batteries, zinc-selenium (Zn-Se) batteries, zinc-tellurium (Zn-Te) batteries, zinc-iodine (Zn-I2 ) batteries, and zinc-bromine (Zn-Br2 ) batteries. Meanwhile, the reaction conversion mechanism of zinc-based batteries with conversion mechanism and the research progress in the investigation of composite cathode, zinc anode materials, and selection of electrolytes are systematically introduced. Finally, this review comprehensively describes the prospects and outlook of aqueous zinc-ion batteries with conversion mechanism, aiming to promote the rapid development of aqueous zinc-based batteries.

RESUMO

Macrophages play a pivotal role in controlling Mycobacterium infection, and the pathogen thrives in the event of immune evasion and immunosuppression of macrophages. Mammalian cell entry proteins (Mce) are required for Mycobacterium tuberculosis (M. tb) growth and the host cell's initial phagocytosis and cytokine response. Mce2D protein is one of a family of proteins that infect M. tb; however, the function and mechanism of action remain unclear. In this study, we constructed the Mce2D knockout strain using Mycobacterium smegmatis to study the function of Mce2D in the infection of macrophages. The results indicated that compared to the knockout strain, the release of proinflammatory cytokines (TNF-α and IL-1ß) reduced when WT strain infected the macrophages. Moreover, Mce2D boosted the metabolism of oxidized fatty acids, increased the energy supply of TCA, and lowered the glycolysis of glucose in macrophages after bacterial infection, all of which prevented the polarization of macrophages to M1, which was driven by the fact that Mce2D blocked ERK2 phosphorylation by interacting with ERK2 through its DEF motif. This, in turn, promoted nuclear translocation of HIF-1α, allowing signal accumulation, which increased the HIF-1α transcription levels. Finally, the mouse infection experiment showed that Mce2D caused blockage of M1 polarization of alveolar macrophages, resulting in reduced bactericidal activity and antigen presentation, weakening Th1 cell-mediated immune response and helping bacteria escape the immune system. Our results reveal that Mce2D causes immune escape by blocking M1 polarization in macrophages, providing potential targets for the rational design of therapies against M. tb infection.

RESUMO

Capacitive deionization (CDI) is perceived as a promising technology for freshwater production owing to its environmentally friendly nature and low energy consumption. To date, the development of high-performance electrode materials represents the foremost challenge for CDI technology. In this work, the porous bismuthene/MXene (P-Bi-ene/MXene) heterostructure was synthesized using a simple interfacial self-assembly method with two-dimensional (2D) bismuthene and Ti3C2Tx MXene. Within the P-Bi-ene/MXene heterostructure, the porous structure can increase the active site and facilitate ion transport. Simultaneously, MXene effectively enhances the conductivity of the heterostructure, resulting in accelerating electron transport. Due to these attributes, the P-Bi-ene/MXene heterostructure demonstrates high desalination capacity (90.0 mg/g), fast desalination rate, and good cycling performance. The simple self-assembly strategy between 2D/2D materials described herein may offer inspirations for the synthesis of innovative electrode materials with high performance.

RESUMO

Aqueous zinc-ion batteries (AZIBs) are considered to be one of the most promising devices for large-scale energy storage systems owing to their high theoretical capacity, environmental friendliness, and safety. However, the ionic intercalation or surface redox mechanisms in conventional cathode materials generally result in unsatisfactory capacities. Conversion-type aqueous zinc-tellurium (Zn-Te) batteries have recently gained widespread attention owing to their high theoretical specific capacities. However, it remains an enormous challenge to improve the slow kinetics of the aqueous Zn-Te batteries. Here, MoO2 nanoclusters embedded in hierarchical nitrogen-doped carbon nanoflower (MoO2 /NC) hosts are successfully synthesized and loaded with Te in aqueous Zn-Te batteries. Benefitting from the highly dispersed MoO2 nanoclusters and hierarchical nanoflower structure with a large specific surface area, the electrochemical kinetics of the Te redox reaction are significantly improved. As a result, the Te-MoO2 /NC electrode exhibits superior cycling stability and a high specific capacity of 493 mAh g-1 at 0.1 A g-1 . Meanwhile, the conversion mechanism is systematically explored using a variety of ex situ characterization methods. Therefore, this study provides a novel approach for enhancing the kinetics of the Te redox reaction in aqueous Zn-Te batteries.

RESUMO

Aqueous zinc ion batteries (AZIBs) have gained extensive attention due to the numerous advantages of zinc, such as low redox potential, high abundance, low cost as well as high theoretical specific capacity. However, the development of AZIBs is still hampered due to the lack of suitable cathodes. In this work, the freestanding defective ammonium vanadate@MXene (d-NVO@MXene) hybrid film was synthesized by simple vacuum filtration strategy. Due to the presence of the hierarchical freestanding structure, outstanding MXene conductive networks and abundant oxygen vacancy (in the d-NVO nanoribbons), the d-NVO@MXene hybrid film can not only expose more active sites but also possess outstanding conductivity and kinetics of charge transfer/ion diffusion. When the d-NVO@MXene hybrid film was directly used as the cathode, it displayed a high specific capacity of 498 mAh/g at 0.5 A/g and superior cycling stability performance with near 100 % coulomb efficiency. Furthermore, the corresponding storage mechanism was elucidated by ex situ various characterizations. This work provides new ideas for the development of freestanding vanadium-based cathode materials for AZIBs.

RESUMO

BACKGROUND: Transmissible gastroenteritis virus (TGEV), which belongs to the coronaviruses (CoVs), causes diarrhea and high mortality rates in piglets and poses a huge threat and loss to the pig industry in China. METHOD: We estimated the prevalence of TGEV in Chinese pig animals from 1983 to 2022 by screening 36 papers on TGEV investigations in China from databases such as China Knowledge Network (CNKI), Wanfang Database, Science and Technology Journal Repository (VIP), PubMed, and ScienceDirect, excluding duplicate literature and other host studies according to the exclusion criteria we developed, and excluding literature with incomplete data to extract information from studies that could estimate the prevalence of TGEV infection in pigs in mainland China. RESULTS: A total of 36 studies (including data from 50,403 pigs) met our evaluation criteria. The overall estimated prevalence of TGEV infection in pigs in China is 10% (3887/50403), and the prevalence of TGEV in northeast China is 38% (2582/3078700) is significantly higher than the rest of China. The prevalence of TGEV infection was related to the sampling season and region. CONCLUSION: The results of the study show that the prevalence of TGEV is clearly seasonal and regional. Therefore, further research and monitoring of the prevalence of TGEV infection and the development of control programs based on different conditions are essential. In addition, effective and robust regulatory measures should be taken in colder regions to prevent the spread and transmission of TGEV in pigs.

RESUMO

Background: Bovine mastitis is one of the most common and prevalent diseases affecting dairy cattle worldwide. It adversely affects the quality and quantity of milk production and leads to a significant economic loss for the farmers. Methods: This article aimed to estimate the prevalence of clinical mastitis (CM) infection in mainland China using a systematic review and meta-analysis. The research reports published during 1983-2022 in English or Chinese from databases (PubMed, Google Scholar, Cochrane Library, Science Direct, Web of Science, VIP Database for Chinese Technical Periodicals (VIP), Chinese National Knowledge Infrastructure (CNKI), and Wan Fang database) were identified after reviewing the relevant scientific literature. Based on our inclusion criteria, this study analyzed the prevalence of CM in 47 published studies prevalence extracted the total number of cattle infected with CM from the available studies, allowing us to estimate the prevalence of CM infection among these in mainland China. Results: The pooled prevalence with the 95% CI for the clinical mastitis was 10% (95% CI: 9.00, 12.00). The majority of CM was associated with lactation, parity, and age, with higher prevalence observed in late lactation 15% (95% CI: 11.00, 18.00) and mid-lactation 10% (95% CI: 6.00, 13.00) in comparison to early lactation 8% (95% CI: 5.00, 10.00). The incidence of CM increased significantly with the increase of parity and age, and the highest incidence rates were 19% (95% CI: 15.00, 23.00) and 16% (95% CI: 12.00, 19.00) at parity and age ≥7, respectively. Among the seasons, the highest prevalence of CM infection was found in autumn 9% (95% CI: 2.00, 17.00). Interestingly, no significant effects were evident regarding the influence of quarter on the prevalence of CM. Conclusion: Thus, estimating the prevalence of CM among cattle in mainland China. through meta-analysis can provide adequate measures to control CM, reduce economic losses, and prevent the spread and transmission of CM in Chinese herds.

RESUMO

Capacitive deionization (CDI) is regarded as a promising desalination technology owing to its low cost and environmental friendliness. However, the lack of high-performance electrode materials remains a challenge in CDI. Herein, the hierarchical bismuth-embedded carbon (Bi@C) hybrid with strong interface coupling was prepared through facile solvothermal and annealing strategy. The hierarchical structure with strong interface coupling between the bismuth and carbon matrix afforded abundant active sites for chloridion (Cl-) capture, improved electrons/ions transfer and the stability of the Bi@C hybrid. As a result of these advantages, the Bi@C hybrid showed a high salt adsorption capacity (75.3 mg/g under 1.2 V), salt adsorption rate and good stability, making it a promising electrode material for CDI. Furthermore, the desalination mechanism of the Bi@C hybrid was elucidated through various characterizations. Therefore, this work provides valuable insights for the design of high-performance bismuth-based electrode materials for CDI.

RESUMO

The distributed acoustic sensing system can obtain the vibration signal caused by the vibration of the train. By analyzing these wheel-rail vibration signals, an abnormal wheel-rail relationship identification scheme is proposed. The variational mode decomposition is employed for signal decomposition, thereby obtaining intrinsic mode functions with prominent abnormal fluctuations. The kurtosis value of each intrinsic mode function is calculated, which is compared with the threshold value for the identification of trains with abnormal wheel-rail relationship. And the extreme point of the abnormal intrinsic mode function is used to locate the bogie with an abnormal wheel-rail relationship. Experimental demonstration verifies that the proposed scheme can identify the train and locate the bogie with an abnormal wheel-rail relationship.

RESUMO

Comprehensive screening for functional substances from natural resources is always a hot research topic. Eicosapentaenoic acid- (EPA) and docosahexaenoic acid (DHA)-structured phospholipids (PLEPA/DHA) have versatile cardiovascular benefits as well as superior bioavailability. Herein, the abundance of PLEPA/DHA in 16 aquatic products was specifically and selectively screened using a recently developed precursor ion scan-driven hydrophilic interaction chromatography-mass spectrometry (PreIS-HILIC/MS) method with the fatty acyl moieties of EPA (m/z 301.6) and DHA (m/z 327.6) locked. The aim focused on the characteristics and differences in the varieties and contents of EPA/DHA-structured phosphatidylcholine (PCEPA/DHA) and EPA/DHA-structured phosphatidylethanolamine (PEEPA/DHA) molecular species. A total of 80 PLEPA/DHA molecules were identified in these natural sources, including 47 PCEPA/DHA and 33 PEEPA/DHA. After analysis, PC 16:0/20:5 and PC 16:0/22:6 are present in all aquatic products and at high levels. Antarctic krill was found to be the best resource of PLEPA/DHA in total (2574.69 µg·g-1), followed by mackerel (2330.11 µg·g-1), salmon (2109.91 µg·g-1), and Farrer's scallop (1883.59 µg·g-1), while abalone contained the lowest level of PLEPA/DHA (310.44 µg·g-1). Besides, sea cucumber and sea bass contained the highest contents of EPA-structured and DHA-structured ether phospholipids, respectively, which could be highly recommended as dietary sources of special functional phospholipids. Finally, the multiple discrepancies between the 16 aquatic products were revealed by multivariate statistical analysis. These findings improve the awareness of the composition and content of PLEPA/DHA contained in aquatic products, providing a reference for their integrated development.

RESUMO

Aqueous zinc-ion batteries have attracted more and more attention due to their safety, environmental benignity and high theoretical capacity. However, the lack of appropriate cathode materials with high capacity and long cycle life have become an obstacle to the development of aqueous zinc-ion batteries. Herein, the hierarchical amorphous vanadium oxide and carbon nanotubes (a-V2O5@CNTs) microspheres with strong interface interaction were successfully prepared by combing facile spray drying technique with annealing treatment. Benefiting from the a-V2O5 amorphous characters, CNTs framework high conductivity and hierarchical microspheres with strong interface interaction, the a-V2O5@CNTs exhibited abundant active sites, fast reaction kinetics as well as eminent structure stability. As a promising electrode material, the a-V2O5@CNTs displayed high specific capacity (480 mAh g-1 at 0.5 A g-1), good rate capability and long-term stability under high current density (158 mAh g-1 at 30 A g-1 over 1000 cycles). Meanwhile, the corresponding mechanism was further illustrated through different characterizations. Furthermore, the as-assembled flexible pouch battery based on the a-V2O5@CNTs delivered outstanding flexibility and feasibility. Hence, this work provides a new idea for developing high performance cathode materials of aqueous zinc-ion batteries.

RESUMO

More and more individuals are paying attention to the research on the emotional information found in micro-blog comments. TEXTCNN is growing rapidly in the short text space. However, because the training model of TEXTCNN model itself is not very extensible and interpretable, it is difficult to quantify and evaluate the relative importance of features and themselves. At the same time, word embedding can't solve the problem of polysemy at one time. This research suggests a microblog sentiment analysis method based on TEXTCNN and Bayes that addresses this flaw. First, the word embedding vector is obtained by word2vec tool, and based on the word vector, the ELMo word vector integrating contextual features and different semantic features is generated by ELMo model. Second, the local features of ELMo word vector are extracted from multiple angles by using the convolution layer and pooling layer of TEXTCNN model. Finally, the training task of emotion data classification is completed by combining Bayes classifier. On the Stanford Sentiment Classification Corpus data set SST (Stanford Sentiment Classification Corpus Data bank), the experimental findings demonstrate that the model in this paper is compared with TEXTCNN, LSTM, and LSTM-TEXTCNN models. The Accuracy, Precision, Recall, and F1-score of the experimental results of this research have all greatly increased. Their values are respectively 0.9813, 0.9821, 0.9804 and 0.9812, which are superior to other comparison models and can be effectively used for emotional accurate analysis and identification of events in microblog emotion analysis.

RESUMO

During spinal cord injury (SCI), the homeostasis of the cellular microenvironment in the injured area is seriously disrupted, which makes it extremely difficult for injured neurons with regenerative ability to repair, emphasizing the importance of restoring the cellular microenvironment at the injury site. Neurons interact closely with other nerve cells in the central nervous system (CNS) and regulate these cells. However, the specific mechanisms by which neurons modulate the cellular microenvironment remain unclear. Exosomes were isolated from the primary neurons, and their effects on astrocytes, microglia, oligodendrocyte progenitor cells (OPCs), neurons, and neural stem cells were investigated by quantifying the expression of related proteins and mRNA. A mouse SCI model was established, and neuron-derived exosomes were injected into the mice by the caudal vein to observe the recovery of motor function in mice and the changes in the nerve cells in the lesion area. Neuron-derived exosomes could reverse the activation of microglia and astrocytes and promote the maturation of OPCs in vivo and in vitro. In addition, neuron-derived exosomes promoted neurite outgrowth of neurons and the differentiation of neural stem cells into neurons. Moreover, our experiments showed that neuron-derived exosomes enhanced motor function recovery and nerve regeneration in mice with SCI. Our findings highlight that neuron-derived exosomes could promote the repair of the injured spinal cord by regulating the cellular microenvironment of neurons and could be a promising treatment for spinal cord injury.

RESUMO

Vanadium-based oxides have gained widespread attention as promising cathode materials for aqueous zinc-ion batteries (AZIBs) due to their abundant valences, high theoretical capacity and low cost. However, the intrinsic sluggish kinetics and unsatisfactory conductivity have severely hampered their further development. Herein, a facile and effective defect engineering strategy was developed at room temperature to prepare the defective (NH4)2V10O25·8H2O (d-NHVO) nanoribbon with plenty of oxygen vacancies. Owing to the introduction of oxygen vacancies, the d-NHVO nanoribbon possessed more active sites, excellent electronic conductivity and fast ion diffusion kinetics. Benefiting from these advantages, the d-NHVO nanoribbon as an aqueous zinc-ion battery cathode material exhibited superior specific capacity (512 mAh g-1 at 0.3 A g-1), excellent rate capability and long-term cycle performance. Simultaneously, the storage mechanism of the d-NHVO nanoribbon was clarified via comprehensive characterizations. Furthermore, the pouch battery based on the d-NHVO nanoribbon was fabricated and presented eminent flexibility and feasibility. This work provides a novel thought for simple and efficient development of high- performance vanadium-based oxides cathode materials for AZIBs.

RESUMO

Capacitive deionization has been considered as a promising solution to the challenge of freshwater shortage due to its high efficiency, low environmental footprint, and low energy consumption. However, developing advanced electrode materials to improve capacitive deionization performance remains a challenge. Herein, the hierarchical bismuthene nanosheets (Bi-ene NSs)@MXene heterostructure was successfully prepared by combining the Lewis acidic molten salt etching and the galvanic replacement reaction, which achieves the effective utilization of the molten salt etching byproducts (residual copper). The vertically aligned bismuthene nanosheets array evenly in situ grown on the surface of MXene, which not only facilitate ion and electron transport as well as offer abundant active sites but also provide strong interfacial interaction between bismuthene and MXene. Benefiting from the above advantages, the Bi-ene NSs@MXene heterostructure as a promising capacitive deionization electrode material exhibits high desalination capacity (88.2 mg/g at 1.2 V), fast desalination rate, and good long-term cycling performance. Moreover, the mechanisms involved were elaborated by systematical characterizations and density functional theory calculations. This work provides inspirations for the preparation of MXene-based heterostructures and their application for capacitive deionization.
